Rat liver polysomes have been found to be unaffected by treatment with p-chloromercuribenzoate under relatively mild conditions. However, in the presence of small amounts of crude enzyme fractions, PCMB brings about a rapid breakdown of polysomes into fractions which consist largely of dimers and monomeric ribosomes. The rapid breakdown of polysomes is due to the action of a ribonuclease enzyme present in the transfer enzyme fraction. Normally, the ribonuclease is in an inactive (inhibited) form. p-Chloromercuribenzoate activates the inactive ribonuclease, thus enabling it to act on polysomes with subsequent breakdown. p.Chlo� romercuribenzoate has no effect on [14C] labeled peptide chains attached to polysomes. The puromycin catalysed-release of labeled peptide chains from polysomes is influenced to a small extent by the presence of PCMB.
